PROJECT
My lab develops engineered T cell therapies for cancer treatment. We focus on understanding and combating cancer cell resistance to CAR-T cell therapy. We have established a high-throughput CRISPR platform that allows for the functional characterization of tumor antigen variants, and we use the newest gene editing technologies to engineer CAR-T cells with improved resilience.

TUM PREP PARTICIPANTS TASKS
- production of CAR-T cells
- transduction of tumor cells with base editing libraries to install antigen variants in the endogenous locus
- set up positive selection screens using tumor cells that express antigen variants and CAR-T cells
- gDNA extraction form screen co-culture and library preparation for next-generation sequencing
- analysis of screening results

EXPECTED OUTCOMES
We expect to find tumor antigen variants that confer resistance to CAR-T cell therapy. Insights from this work will inform engineering efforts to make CAR-T cells less vulnerable to tumor resistance.
