Project description:

Genetically encoded biosensors are our window into the distributions of small molecules and ions in cells and tissues in vivo. While such tools are already well established for imaging of cells by fluorescence microscopy they are largely lacking for the interrogation of molecular distributions in whole opaque organisms. Here we develop innovative sensor concepts for such imaging schemes. For this we use techniques of protein engineering encompassing mutagenesis, directed evolution and screening. Further, we characterize sensors on their photo-physical functioning levels as well as structural details. Finally, we seek to demonstrate sensor function in proof of principle applications.
